Protocols

  • Slow escalation to 9 mgrecommended

Reproduces the 3/6/9 mg arm of NCT06230523. Investigational; Monday dosing.

  • Faster escalation to 9 mg

Reproduces the 6/9 mg arm of NCT06230523. This arm had the highest reported nausea and fatigue rates among the escalation arms.

Dosing

  • Trial arm 3/6/9 mg: 3 mg once weekly for 4 weeks, 6 mg for 4 weeks, then 9 mg for 40 weeks (48 weeks total).
  • Trial arm 6/9 mg: 6 mg once weekly for 20 weeks, then 9 mg for 28 weeks (48 weeks total).
  • Other trial arms were fixed 1, 3, 6 or 9 mg once weekly for 48 weeks.

Route & timing

Route: Subcutaneous, once weekly (trial).

Timing: Same day each week; the sources give no time-of-day guidance.

Reconstitution

  • No reconstitution figure exists: the trial product is a Lilly clinical-supply formulation, not a research vial. The water in the protocols is arithmetic for a readable draw, not a sourced instruction.

Stability

No reliable figure found in the sources; ask your prescriber or supplier.

Off-cycle

No off-cycle in the trial (continuous 48 weeks).

Why:

  • Trial arms were continuous; no sourced washout.

Stacking rules

  • Never draw into the same syringe as another compound; use separate syringes and distinct injection sites.

Safety

Investigational; not approved anywhere. Any vial sold outside a trial is unapproved and unverified.

Common side effects:

  • Most common in the phase 2 trial: nausea (up to 64% at 6 mg, 33% at 9 mg vs 14% placebo) and fatigue (up to 46% in the 6/9 mg arm), mostly dose-related (PubMed abstract).
  • Per Outliyr: constipation 24%, vomiting 11% and alopecia 11% at 9 mg; slow heart rate in 11% of the 6 mg arm (unexplained).

Stop and call your prescriber if:

  • Severe or persistent vomiting, or signs of dehydration
  • Severe upper abdominal pain radiating to the back
  • Resting heart rate well below normal, or dizziness on standing
  • Shakiness, sweating or confusion, especially with insulin or a sulfonylurea
